Mapping

Kissebah et al. (2000) performed a genomewide scan by use of a 10-cM map in 2,209 individuals distributed over 507 nuclear Caucasian families. Pedigree-based analysis using a variance components linkage model demonstrated a QTL on chromosome 17p12 strongly linked to plasma leptin levels (lod = 5.0). This QTL has possible epistatic interaction with another QTL on chromosome 3q27 (605552) that is strongly linked to 6 traits (weight, waist circumference, leptin, insulin, insulin/glucose ratio, and hip circumference) associated with the phenotype (lod scores ranging from 2.4 to 3.5). Several candidate genes are located in both regions.
